LabMidterm Flashcards
Two precautions
- No eating/drinking
- Labcoat
- Wash hands frequently
Phase contrast microscopy useful for viewing
wet mounts (unstained slides)
When streaking agar solution… Failing to loop would result in..
No isolated colonies.
Primary stain
Crystal violet
Gram + and Gram - are purple…
Mordant
Gram’s iodine
Gram + and Gram - are purple…
Decolourizer
Acetone - Alcohol
Gram + (purple)
Gram - (clear, colourless)
Counterstain
Safranin
Gram + (purple)
Gram - (pink)
What step in gram stain process makes stain differential?
Decolourizer - acetone-alcohol. It rapidly decolourize Gram - bacteria and gram+ will remain purple. Thin wall of negative allows the iodine to wash out easily
Positive string test indicates..
Gram negative cell wall
Antibiotics are natural chemicals.. explain
Antibiotics are made from microorganisms and are therefore natural.. They are not synthetic (created in a lab)
Factors that will affect the zone of inhibition with disc with disinfectant..
- type of bacteria/bacterial resistance
- concentration of disinfectant in disc
- pH of disinfectant
Physical conditions necessary for sterilization in autoclave
15 psi, 121degrees for 20 mins
Method used to ensure proper operation of autoclave
autoclave tape or strips with heat sensitive dye used. when proper conditions reached - invisible dye becomes visible.
Choice of sterilizing a flask of culture medium?
autoclaving
choice of sterilization for plastic petri dishes?
gamma radiation
