8
Q

What step in gram stain process makes stain differential?

A

Decolourizer - acetone-alcohol. It rapidly decolourize Gram - bacteria and gram+ will remain purple. Thin wall of negative allows the iodine to wash out easily

16
Q

Chioce of sterilization for liquid?

A

filter sterilization

17
Q

Why agar side up?

A

So nothing drips from the top onto the experiment being carried out. Ex - condensation won’t collect on agar, but on lid instead.

18
Q

Well streaked if…

A

individual colonies

19
Q

punctiform

A

small dots.. <1mm dia

20
Q

entire

A

whole margin

21
Q

undulate

A

wonky line

22
Q

filamentous

A

bundles

23
Q

bascillus

A

Bacillus is a genus of Gram-positive, rod-shaped bacteria

24
Q

String test

A

KOH added, cell walls break down if negative and dna strings behind a loop as lifted. In positive, cell walls fail to break down in KOH

25
Q

structural stains

A

used to identify specific parts of microorganisms. Help to identify spores or capsules etc

26
Q

Capsules

A

slimy gelatinous layer of material surroundings bacterial cell.

  • can’t be stained. Halo effect
  • not heat fixed prior to staining because capsule would be affected and results inconclusive
27
Q

four ways to adjusting illumination on light microscope

A
  • condenser diaphragm
  • condenser
  • light intensity adjustment
  • phase contrast microscopy option
28
Q

peptidoglycan

A

structure in cell walls

29
Q

boiling

A

does not kill endospores
