Flashcards in Pharmacokinetic Principles Deck (19):
1

What is Pharmacokinetics?

  • Study of the time course of drug in the body and the relationships between this time course and the dose given

2

What is Pharmacodynamics?

  • The effects of a drug on the body and the relationships between dose/concentration and pharmalogical response

3

What is Biopharmaceutics?

  • Factors which influence the rate and amount of drug reaching the systemic circulation

4

What Parameters are CL, Vd and F?

  • Primary pharmacokinetic parameters
    • have their own unique set of determinants which are specific to the drug [physicochemical properties] and the physiology, biology and biochemistry of the human body

5

What Parameters are k and t1/2?

What are they determined by?

  • Secondary parameters
  • Secondary parameters are determined by primary pharmacokinetic parameters
    • Secondary parameters do not determine primary parameters.

6

What is Absorption?

  • Input into the systemic circulation from extravascular administration site

7

What is Distribution?

  • Reversible movement of drug between the systemic circulation and other organs and tissues of the body

8

What is Excretion?

  • Movement of drug into excretory fluids, physical removal of drugs

9

What is Elimination?

  • Irreversible loss of drug from the body by excretion or metabolism

10

What is Disposition?

  • Drug distribution and elimination - all processes that occur after absorption

11

What is Clearance?

  • The efficiency of drug elimination from the body

12

What is Bioavailability?

  • Fraction of an extravascular dose which actually reaches the systemic circulation intact

13

What is Volume of Distribution?

  • Measure of drug distribution throughout the body

14

What is Extraction?

  • Ability of an eliminating organ to extract drug from blood/plasma into metabolite or urine

15

What is Elimination Rate Constant?

  • k
  • First order rate constant describing the first order elimination of a drug

16

What is Elimination Half-Life?

  • The time taken for 50% of the drug to be eliminated from the body

17

Pharmacokinetic Date: Define Input

  • Rate and extent of entry to systemic circulation

18

Pharmacokinetic Date: Define Distribution

  • Loss from and return to Plasma

19

Pharmacokinetic Date: Define Output

  • Parameters reflecting net removal - elimination
