Name some general properties that NSAIDs have in common.

A
  • All but one are weak organic acids (nabumetone, a ketone prodrug that is metabolized to the acidic active drug)
  • most are well absorbed, food does not sign. change their bioavailability
  • highly metabolized (largely by liver, cyp3A or cyp2c families of P450 enzymes)
  • Renal excretion is the most important final route, but nearly all undergo some degree of bilirary excretion & reabsorption
  • most highly protein bound, usually to albumin
  • most are racemic mixtures
  • all can be found in synovial fluid after repeated dosing
