What is the pronciple of chromatography?
U or I could Kiss! 🌚
Chromatography is an analytical technique used for separating mixtures of chemical substances into individual components which could be further analysed
Principle of chromatography is based on 2 ideas
Solibility
Adsorption
Explain how
TOTT
TOTI
GR- OT- CIT - TT- RIT
Adsorbtion
The Ability of a compd to attach to the stationary phase
Solubility
The ability of a compd to dissolve in the mobile phase
These properties gives rise to the different affinity of the individual components contained in the analyte towards the mobile and stationary phase resulting in the differential separation of the compds.
Relationship b/w adsorption and elution?
The greater the adsorption of components to the stationary phase the slower the elution of components
The greater solubility in mobile phase the faster elution of component from th me column
Affinity of compd to mobile or stationary phase is based on polarity of components
2 classes of chromatography?
Normal phase chroma
Reversed phase chroma
The stationary phase is polar
Mibile phase non polar.
Reversed is opposite if above said …. Eg HPLC
Priciples and methods
Paper
Thin layer
Liquid
Size exclusion
Gas chroma
Polarity of compd(1,2,3)!!
Based on size of particles
Based on boiling point
Spotting on celluloce
Spotting on TLC paper
Glass column packed w slurry silica
Large particles are eluted first b4 smaller particles
The components w low boiling point first b4 H.BP
4 other types of chromatography
SAID
Supercritical chroma
Ion exchange chroma
Affinity chroma
Displacement chroma
