Avoids extrem pH and osmotic pressure and high temperature; employs aqueous solution; Temperature= O-4degrees Celsius; to avoid loss of biologic activity
(1) Extraction
Distrups the cell to liberate its contituents; resulting suspension contains intact organelles known as homogenate

Manually operated or motor driven pestle is rotated within a glass tube containing the minced fragments of the organ under and a suitable medium

Done at successively greater speed, each yield a pellet and supernatant
