Q

EMB

A

eosin methylene blue agar - contain lactose + differentiated between lactose fermenters based on their acid production. differential for type of g- but selective for gram negative too becasue dye will kill g+ bacteria

Q

bile-esculin test

A

capable of growth in in presence of bile _ esculin. glycoside found in several plants.
esculin hydrolysis = glucose. esculetin + ferric salt = dark brown-black colour. colour change =+.

Q

BAP test

A

exoenyme = hemolysins rupture rbc membrane. ability to perform hemolysis = key test.
a-hemolysis: formation of greenish zones - oxidation of iron atom due to H2O2. not true.
b-hemolysis: clearing around colonies due to lysis of rbc = true hemolysis
y-hemolysis: no clearing, dicoloration.

Q

oxidative test

A

differentiate enteric g- acteria. test for presence of cyt c oxidase. = essential to obligate aerobes. reduced TMPD (-) for cyt c = purple. TMPD oxidied = cyt c + = blue rapidly.

Q

EMB agar

A

selective and differential medium contains dye to inhibit g+. black metallic = e coli. mixed acid fermentation. butanedial fermentation = e. aerogenes. fish-eye.
lactose non-frmenter: pale/colourless proteus vulgaris.

17
Q

Macconkeys

A

selective contains crystal violet to inhibit g+. contains NaCl and bile to inhibit non-enteric bacteria.
lactose fermenter => acid + neutral pH = red/pink color to colony. e.coli crisp red. e aerogenes pink, gelatinous
