What is the theory being the Verhoeff Elastin Stain?

A

Tissue overstained with iodine-ferric chloride-hematoxylin solution. Ferric chloride is mordant and differentiator, iodine oxidizes hematoxylin and elastin. Elastin is the last element to be decolorized due to strong affinity. Iodine is removed by sodium thiosulfate.

What is the theory behind Gomori’s Aldehyde Fuchsin?

A

Aldehyde fuchsin ripens (2-4 days), used progressively to demonstrate elastin. Oxidation with potassium permanganate will intensify staining, not required.

Old/Improperly stored Aldehyde fuchsin (store at 4 degrees), Paraldehyde is too old (part of aldehyde fuchsin), slides cut too far in advance. Depending on age of solution, staining times vary.
