Q

What is Cycloheximide chase assay?

A
  1. Add Cycloheximide at point 0
(blocks translation and thus de novo synthesis)
  2. Take samples
  3. Prepare cell extracts
  4. Gel electrophoresis and Western blot

7
Q

What are advantages of Cycloheximide chase?

A

Starts from stady state level but only works if degradation system does not require de novo synthesis of proteins

Q

What is tandem fluorescent protein timer?

A

Detects stability of proteins in vivo
Uses fusion protein. X-GFP-mCherry
Stable proteins have same expression GFP + mCherry
short lived protein much higher GFP

16
Q

Whats autophagy important for?

A

Under starvation conditions

17
Q

Describe the Atg8 + Atg12 pathway in autophagy

A

ATG12 binds to ATG5 and then to the PAS (preautosomal strucure) which then allows ATG8 bound to PE to also bind to the PAS and initiate autophagosome formation

18
Q

What is LIR used for?

A

cargo recognition

19
Q

What is LIR used for?

A

cargo recognition
