Outline the practical procedure of measuring enzyme activity with trypsin and milk

A

-immerse equal volumes of trypsin and milk in a water bath for 5 mins
-mix together and immediately start a timer and record time taken for solution to be fully hydrolysed (becomes colourless)
-test at 5 different temperatures with at least 3 repeats

what is the effect of temperature on enzyme activity

A
  • as temperature increases, kinetic energy increases so more enzyme substrate complexes form so rate of reaction increases
    -past optimum temp, bonds in tertiary structure break and changes shape of active site
    -substrate no longer hydrolysed by denatured enzyme, rate decreases
