● MOA : Acts on amylin receptors in the hindbrain or by a vagally­ mediated mechanism to:
● Slow gastric emptying
● Suppress post­prandial glucagon concentration
● Increases Satiety (i.e., decreases appetite)

What are the characteristics of Pramlintide ( Symlin )?

A

● Type­1/Type­2diabetics who use insulin at mealtime and have failed to achieve diabetic control
● Injected S.C. before meals (reduces post­prandial glycemia)
● Weightloss
● S.E. =
● Nausea (28­-48%)→titrate dose to avoid
● Vomiting, anorexia, headache
● Hypoglycemia
● Caution in patients with diabetic gastroparesis
● Drug interactions because it slows gastric emptying →Don’t take with other drugs within 1 hour before or 2 hours after injection. Don’t co­administer anti­cholinergics, (that may slow gastic motility ) acarbose or miglitol
● Half­-life = 48 minutes
● No hypoglycemia if used alone, but increases risk of severe hypoglycemia if used with insulin
