Why is genotyping wild populations relevant?
To clarify population structure and gene flow, identifying hard to identify species, estimate levels of genetic variability, reconstruction of phylogenetic trees
What is High-Resolution Melting?
A closed tube highly se sitive genotyping alternative capable of Distinguishing single nucleotide polymorphisms(snp) and short insertion/deletion events by examining the melting profile of the PCR products
What is LC green?
A saturation dye
Do homozygotes or heterozygotes have a higher melting temperature?
Homozygotes have a higher melting temp, making them easier to distinguish from heterozygotes
What has high resolution melting analysis been used for?
Forensic identification of marine fauna, avian sex identification, and to genotype loci for population genetic studies
Is sequencing for forensic studies cost effective?
No because it is time consuming
What are the shortcomings of PCR-RFLP
More restriction enzymes are needed, more gels are needed, more lab time, and more reagent and personnel costs

Requires the design and use of multiple oligonucleotide primers and gel electrophoresis
