Explain what is meant by Q10
The initial increase in rate of reaction in a 10 degree Celsius temperature interval
Devise a method for investigating the effect of temperature on the rate of enzyme activity
IV - temp of water bath (10, 20, 30, 40, 50 °C)
DV - gas collected in 20 seconds
CV - vol and conc of solutions (use set moles)
-Keep enzyme and reactant in water bath to acclimatise
- add set volumes to syringes, putting the reactant in the tube and sealing with a bung, and the enzyme pipette in the top
- start the stopwatch whilst adding the enzyme and shake for 10 seconds
- stop shaking and start collecting gas over water between 10-30 seconds
- repeat for each temp to find a mean, plot mean rate on a graph and find Q10 by finding difference in rate in an initial temp change of 10°C
