To make a genomic DNA library

A
  1. chromosomal DNA is extracted.
  2. It is digested with restriction enzymes, and the resulting fragments are inserted into vectors
  3. Vector selection: the library ideally contains the whole genome in the smallest number of clones.
  4. BACs and YACs used to accommodate the large sizes of DNA necessary to span the approximately 3 billion bp of human DNA (as in the Human Genome Project).
