What is the process of Semiconservative replication of DNA?
Semiconcervative replication of DNA is the process in which new DNA molecules replicate off of one original DNA strands becoming a newly synthesised strand and anotherstrand stays original (parental)
What is the DNA Replication Process?
- Helicase enzyme separates the double helix of DNA and slits the DNA into 2 strands by breaking down the hydrogen bonds between the base pairs
- When the strands are separated they can be refered to as the template strands
- DNA polymerase links nucleotides toghether to form theb new strand, using pre-existing strands as a template
What enzyme is the building of the new strand during the replication of DNA carrie by?
The building of the new strand is carried by the enzyme called DNA polymerase, which links nucloetides toghether to form a new strand using the pre-existing strand as a template
How does DNA polymerase move?
DNA polymerase moves a long the template strands in the 5 prime to 3 prime direction, adding one nucleodite at a time
What is PCR?
Its a technique used to amplify section of DNA (used to make many copies of specific segments of DNA)
What is the purppose of DNA replication?
It purpose can be to recoover DNA information from a crime scene or in order to find family connections
What is PCR used for?
Forensic analysis and paternity determination, and DNA found in crime scenes
What are the PCR steps?
- Melting: heat is applied to separate the strands of the double helix of DNA. Heat breaks down hydrogen bonds that hold the strands together. Its a form of measured denaturation
- Annealing: heat is then reduced so that the primers can bind to match the original DNA sequence
- Elogation: heat is added again and taq polymerase enzyme is added to bind to the primer sequence and add nucleotides to extend the second (new stand),

What does the PCR (Polymerase Chain Reaction) do?
It provides a way to make more copies of a portion of DNA
What is taq polymerase?
Its an enzyme used during elogation fase of PCR, to bind with the primer sequence of DNA to extend the sequence and replicate DNA
What is gel electrophoresis and what is it used for?
Gel electrophoresis is used for separating molecules based on size. In DNA it is used to see if DNA copied correctly, to meassure the copies of DNA, and compare DNA samples
What is the primer?
The primer is a chunk of RNA that is attached to each DNA stand marking the starting point for the construction of the new strand of DNA, after Helicase
