DNA structure and history Flashcards
(17 cards)
virulent strain that leads to death in mice
leads to smooth colony
S strain
benign strain (nonvirulent), mice don’t get sick
leads to rough colony
R strain
difference between S and R strains
R lacks protective polysaccharide capsule
what happens when live S is injected into mice in Griffith’s experiment
mice get sick and die
what happens when live R is injected into mice in Griffith’s experiment
mice stay healthy
what happens when heat killed S is injected into mice in Griffith’s experiment
mice stay healthy
what happened when mixture of heat killed S and live R were injected into mice in Griffith’s experiment
mice got sick and died
what did Griffith’s autopsy find in mice who died with heat killed S + live R
blood was full of live S bacteria, no R bacteria
something from heat killed S bacteria got into the live R strain of bacteria and transformed them to virulent S bacteria
most scientists ignored this
Griffith’s conclusion
same as Griffith’s experiment but no mice due to criticism from others who said Griffith’s results were caused by something in the mice
Alloway experiment
Alloway: in test tube, incubated mixture of heat killed S with live R
incubated overnight and checked under microscope next day RESULT
only S bacteria
since transformation of R to S occurred in a test tube without need to inject bacteria into mice and since only thing that is known to have the ability to transform to living things:
live bacteria somehow obtained genetic material form dead S
Alloway’s conclusion
Avery et al: same experiments but no S bacteria
in test tube incubated with DNA extracted from heat killed S with live R
incubated overnight and checked under microscope next day
RESULT
all R bacteria turned into S bacteria
DNA led to transformation of R bacteria into S bacteria
therefore, DNA must be the genetic materia
Avery et al conclusion
when DNA was extracted, wasn’t a clean extraction, may have pulled proteins also
criticism of Avery et al experiment
used to inject phage
bacterial phage
