DNA Structure & Function Flashcards
(12 cards)
Name the DNA bases.
- Adenine
- Cytosine
- Guanine
- Thymine
Which bases are purines and which are pyrimidines?
Purines: A + G, 9-membered rings
Pyrimidines: C + T, 6-membered rings
How is DNA polarity denoted?
DNA has polarity denoted 5’ –> 3’
How are deoxynucleotides linked?
Deoxynucleotides are linked by 3’ –> 5’ phosphodiester bond.
What is the diameter of the helix?
The helix has a diameter of 20 Å.
What distance separates two subsequent bases in the helix?
The bases are separated by 3.4 Å.
How many bases (distance) does the helix structure repeat?
The helix repeats every ~10.5 bases (34 Å).
What are the possible types of DNA helices?
B form (right handed)
- most prevalent form
A form (right handed)
- less prevalent
Z form (left handed)
- not common (transient)
Who discovered DNA was the “transforming principle”?
Avery, MacLeod, McCarthy
Describe the Avery, MacLeod, McCarthy experiment.
Experiment:
What factors influence DNA denaturation (melting)?
higher GC content –> higher Tm
higher [salt] –> higher Tm
longer DNA length –> higher Tm
How do circular chromosomes resolve topological constraints?
They require topoisomerase activity to resolve topological constraints.
