Drugs Flashcards
(18 cards)
Temozolomide
Damage DNA; crosslinking; Alkylating agent
Non-specific
PO or IV
BBB? Yes, need activation
Similar to Cyclophosphamide
Potential Side Effects: Secondary neoplasm, rapidly diving cells prone
Vincristine
Interrupts mitosis; Inhibits microtubule formation
M-phase specific
IV
BBB? Yes
Similar to Paclitaxol
Side effects: Immunosuppression, hair loss, etc…
Carmustine
Alkylating agent (DNA and RNA) Non-specific IV or intracranial wafer BBB? <50% poor Similar to Cyclophosphamide Adverse effects: Infection, meningitis
Cyclophosphamide
Nitrogen mustard alkylating agent
Non-specific
PO, IV
BBB? Yes
Doxorubicin
Intercalates DNA
Non-specific
IV
BBB? No
Methotrexate
Inhibit nucleotide synthesis; Antifolate
S Phase Specific
PO, IV, IM, SQ, intrathecal
BBB? No
Etoposide
Cytotoxic topo II inhibitor (strand breaks)
S-phase specific
IV
BBB? Yes
Ara-C
Prevent DNA synthesis; Inhibit DNA synthesis enzymes; Pyrimidine
Busulfan
Damage DNA; Crosslinking
Cisplatin
Damage DNA; Crosslinking; Platinum
Cyclophosphamide/ifosfamide
Damage DNA; Crosslinking; Alkylating agent
Fludarabine
Prevent DNA synthesis; Inhibit DNA synthesis enzymes; Purine
5-fluorouracil
Prevent DNA synthesis; Inhibit nucleotide synthesis; Pyrimidine
Paclitaxel
Interrupts mitosis; Promotes microtubule formation
Dexamethasone
Immunosuppresant
Reconstitutes the BBB by decreasing swelling
Antiemetic
Crossing the blood brain barrier?????
Small Lipophilic Efflux Pump?
TMZ yes yes nope
BCNU no somewhat meh
Vincristine no yes pumped out
Amphoterecin no yes pumped out
Adverse affects of the Gliadel wafer?
Can cause infection, this can be bad because immune system is weakened by dexamethasone and/or chemo drugs and body can’t fight off on it’s own…..sometimes need to treat with antifungal or antibacterial….
Adverse affects from DNA alkylating agents?
Secondary neoplasms (B cells can be targeted because they divide rapidly)
