Q

Whats ELISA step 1?

A

Antigens from the sample are attached to a surface (usually a multi-well plate),
time is then given for the antigen to adhere to the plate.
A solution of a nonreacting protein (eg bovine serum albumin or powdered milk) is added to the plate to block any unbound sites to prevent non-specific binding of the antibody.

Q

Whats ELISA step 2?

A

A specific antibody is applied to the surface so that it can bind with the antigen.
This antibody is linked to an enzyme,

Q

Wtf is ELISA step 3?

A

a substance containing the enzyme’s substrate is added.
The subsequent reaction produces a detectable signal, most commonly a colour change.
The higher the concentration of antibody, the stronger the colour change, which can then be detected by a spectrometer

Q

What is ELISA? [FULL]

A

is a technique using antibodies and colour change to identify a concentration of an antigen in a mixture. Antigens from the sample are attached to a surface (usually a multi-well plate), time is then given for the antigen to adhere to the plate. A solution of a nonreacting protein (eg bovine serum albumin or powdered milk) is added to the plate to block any unbound sites to prevent non-specific binding of the antibody. A specific antibody is applied to the surface so that it can bind with the antigen. This antibody is linked to an enzyme, and then a substance containing the enzyme’s substrate is added. The subsequent reaction produces a detectable signal, most commonly a colour change. The higher the concentration of antibody, the stronger the colour change, which can then be detected by a spectrometer. Direct or indirect (sandwich) ELISAs can be used (details of indirect ELISA are not required).
