F1. Bacterial smear prep & gram staining Flashcards
(29 cards)
A dried preparation of bacterial cells on a glass slide
Bacterial Smear
Properly processed smear:
- evenly spread out
- bacteria are not washed out during staining
- bacterial form is not distorted [smears are sterile & bacteria are not killed)
Good smears are critical for discerning:
- morphology (shape)
- arrangement (clustering)
- internal structures
two loopfuls of liquid containing organisms from culture
Broth cultures
two loopfuls of water + very small amount of organism [organism should be emulsified w/ the water]
Plates & Slants
use of a single stain to color a bacterial cell
Simple staining
- basic dyes because they have color-bearing ionic groups (chromophores) that are positively charged (cationic)
- commonly used dyes in simple staining:
- methylene blue
- basic fuchsin
- crystal violet
Coccus?
Spherical
four arrangements of coccus shape?
- diplococcus
- streptococcus
- staphylococcus
- micrococcus

streptococcus
random or grapelike clusters
staphylococcus
square groups of four cells

pleomorphic
it has two kinds of cells:
gram positive & gram negative
- stained purple
- stained red/pink
Gram staining
- gram positive
- gram negative
cultures should be ____ hours old…
16-18
critical step in gram staining procedure?
prepare thin smears
most critical step in gram staining procedure?
decolorization
what separates the organism that will be stained
decolorization
