Ka 2 Dna Replication Flashcards

1
Q

What’s the role of DNA polymerase in DNA replication?

A

Adds DNA nucleotides using complimentary base paring to the deoxyribose 3’ end of the new DNA stand which is forming.

Q

Describe the process of DNA replication

A

The DNA unwinds and the hydrogen bonds are broken. Which gives u 2 strands. A primer is needed to start replication.
Leading strand: primer binds to the target strand, then DNA polymerase adds nucleotides to the 3’ end continuously and then forms of a sugar, phosphate backbone between nucleotides. Repeated until entire strand is replicated.
Lagging stand: the strand is replicated is fragments. Joined by Ligase. As DNA separates another fragment will connect until entire molecule is replicated.

Q

Stages in the pcr process

A

•Heat up 92-98c to separate strands
• cooled 50-65c to let primers to bind with target sequence
• heated between 70-80c for heat tolerant DNA polymerase to replicate the region of DNA
• which amplifies the target region of DNA

Q

Role of primer in the PCR process

A

The starting point for DNA replication!
they bind to a specific segment of DNA and signal the DNA polymerase to begin copying the DNA
