Confirmed Test
performed to confirm if any organism is gram negative and produces acid from latose fermentation
Streptococci
Gram +, mostly pathogens, hemolysis test
Alpha
incomplete lysis, green zone
Beta
clearing around colonies due to complete lysis
Gamma
non hemolytic
Staphlyococci Media 110 (SM110)
selective due to high salt conc of 7.5%. clear medium.
Mannitol Salts Agar (MSA)
selective due to high salt conc of 7.5%. differential for mannitol fermentation due to phenol red pH indicator. fermenters turn plate YELLOW due to acid production
Rabbit Plasma
differential for coagulase production. indicated by medium solidifying after inoculation.

electrical pulses make the cell membrane permeable to allow DNA to enter the cell

treat cells with high concentration of MgSo4 and heat shock
storage buffer: polyethylene glycol
acts like antifreeze to keep liquid from freezing

Plasmid Inhibition Control
LB medium with cells and plasmic. Indicated if cells survived transformation and that the addition of pUC18 did not inhibit growth. expect lawn.
resazurin
indicates if oxygen is present (pink) or not (clear)
sodium thioglycolate
reduces free oxygen in the medium to water
fluid thioglycolate medium
broth that allows for growth of anaerobes in aerobic conditions
brewer’s agar
used to streak for growth in anaerobic jar
anaerobic jar
sealed incubation container
gaspak
added to jar to create anaerobic environment. generates H that binds with O2 present to reduce it to H2O
methylene blue strip
