11
Q

how does a TEM work ?

A

TEMs use electromagnets to focus a beam of electrons, which is then transmitted through the specimen. Denser parts of the specimen absorb more electrons, which makes them look darker on the image you end up with.

Q

What is one disadvantage of using a TEM ?

A

Can only be used on
thin specimens. Can only be used on non-living specimens

13
Q

what is one advantage of TEM?

A

Give high resolution images,
so shows small objects.

14
Q

How does a SEM work ?

A

SEMs scan a beam of electrons across the specimen. This knocks off electrons from the specimen, which are gathered in a cathode ray tube to form an
image.

15
Q

what are the disadvantages of using SEM?

A

Give lower resolution
images than TEMs.
Can only be used on non-living specimens.

16
Q

what are the advantages of using SEM?

A

Can be used on thick
specimens. Can be 3-D.

17
Q

what is the first step to preparing a microscope slide ?

A

pipette a small drop of water onto the centre of the slide

18
Q

what is the second step of preparing a microscope slide ?

A

Use tweezers to place a thin section of the specimen onto of the water drop

19
Q

what is the third step of preparing a microscope slide ?

A

Add a drop of a stain.

20
Q

give 2 examples of stain and what they’re used for

A

Eosin is used to make the cytoplasm show up. Iodine in potassium iodide solution is
used to stain starch grains in plant cells.

21
Q

what is the fourth step of preparing a microscope slide ?

A

carefully lower the cover slip down onto the water drop
