a change in the state of neurons or a group of neurons that alters its response to a subsequent stimulation

- commonly act on the release of neurotransmitters from the axon terminals/presynaptic neurons/postsynaptic receptors
- most bind to GPCR therefore act on slower timescale relative to neurotransmitters that bind to ionotropic receptors
- GPCR = metabatropic
- ion channels = ionotropic
- examples: ACh, dopamine, seratonin, cannabinoids, nitric oxide
