Define Amino acids:
molecules containing an amine group, a carboxylic acid group and a side-chain that varies with different amino acids
What are peptides?
short polymers of amino acids linked by peptide/amide bonds
What are the reactants for dipeptide synthesis?
Alanine methyl ester and Boc phenylalanine
What are the reagents for dipeptide synthesis?
EDCl and DMAP
What is the solvent?
Dichloromethane
What groups are protected?
- NH2- of phenylalanine protected as t-butoxycarbonyl
- COOH of alanine is protected as methyl ester
What is DMAP used for?
acts as base/activator
What is EDCl used for?
activation of the carboxylic group
What is HCl washing used for?
remove unreacted amine, DMAP (sm)
What is NaHCO3 washing used for?
Remove unreacted acid (sm)
What is water washing for?
remove urea, any other salts (byproducts of reaction)
What is brine washing for?
remove excess water
Whats the purpose of sodium sulfate?
remove trace water
Where to dispose methylene chloride?
liquid organic hazardous waste
Where to dispose all aqueous waste generated during extraction?
hazardous aqueous liquid acidic and basic chemical waste
What are the layers?
Top aqueous bottom organic (DCM)
What is the stationary and mobile phase in TLC?
Stationary: silicon dioxide
Mobile: 10% methanol in chloroform
