Type of mount where the specimen is mounted on the slide with our without coverslip. It is quick and easy to make but it dries out live sample.
Dry Mount
Type of mount where the specimen is mounted on the slide with 1-3 drops of water, with coverslip. It is useful for viewing cellular movement, aquatic organisms, etc.
Wet Mount
Type of mount where the specimen from a semiviscous/viscous medium is spread thin across the slide. Allows cells to be viewed
separately, distinguish
cellular debris, organic and
non-organic matter
Smear Slide
Type of mount where the specimen is thinly sliced
using a blade
Free-Hand Sectioning
Type of mount where the specimen sections are
arranged in sequential
order and were cut using a
microtome
Serial Sectioning
Type of mount where a thinly-sliced soft specimen is
placed on the slide and
gently pressed down by the
coverslip. Useful for viewing minuteparts of a macroscopic
structure
Squash Slide
Fixating reagent used
Farmer’s fluid (3:1 ethanol to acetic acid)
Reagent used to further fix the sample if it is not to be used immediately
70% aclohol
Reagent used to dissolve the pectic substance of the middle lamella
1N HCl
Reagent that softens and swell tissues, making it easier to spread cells or chromosomes on slides. Also acts as a fixative
45% Glacial Acetic Acid
Staining reagent for nucleus/chromosomes
Acetocarmine
Seals the cover slip on the slide
Paraffin
