Prostoglandins (PGs) Flashcards
MoA of NSAIDs
inhibition of cyclooxygenase (COX) enzymes.
* Reduction in eicosanoid formation from arachidonic acid
→ ↓ prostaglandins synthesis (↑ eukotrienes synthesis)
Non-inflmatory drugs include?
1) NSAIDs
2) Glucocorticoids
MoA of PGE2
prominant in inflammatory respone –> mediator of fever and pain
- Inhibition of gasteric acid secretion
- Relaxation if broncial, vascular and GI tract SM
MoA of PGI1 (Prostacyclin)
vasodilatation and inhibition of platelet aggregation
MoA of PGF2a
1) contraction of the uterus
2) decreases intraocular pressure
MoA of PGD2
vasodilatation and inhibition of platelet aggregation
category of Misoprostol
PGE1
indication of Misoprostol
protective action on gastric mucosa
MoA of Misoprostol
Prostaglandin
receptor
agonists.
PK/PD of Misoprostol
- Misoprostol –> a metabolically stable
prostaglandin (PG)E analogue.
- It has the potential risk to induce
abortion in pregnant women à therefore,
the drug is contraindicated during
pregnancy
Adverse effects of Misoprostol
Contraindicated In pregnancy,
unless –> used as an abortifacient(misoprostol in combination with mifepristone)
indication of Alprostadil
- Erectile dysfunction.
- Maintain ductus arteriosus potency
MoA of Aloprostadil
Prostaglandin receptor agonists.
PK/PD of Alprostadil
-
Vasodilation –> for impotent males to
treat erectile dysfunction. -
Maintains potency of ductus arteriosus
in neonates until surgical correction in
neonates with certain congenital heat
malformations
Indication of Dinoprostone
-
“cervical ripening”.
(Induction of labor) - As abortifacient
