What is a plasmid?
Bacterial DNA that can be used to insert a gene
What is the antibiotic resistance gene used for?
It is used to select for the gene of interest so, when you spread the bacteria with the plasmid the only bacteria that will survive are those that have the resistance gene and those will also have you gene of interest
How do we obtain plasmid DNA?
- Grow E. Coli in culture with the antibiotic to select
- Heat shock treatment
What is transient gene expression?
A short term gene expression that is not inherited
What does nanodrop spectrophotometry do?
Quantifies the yield concentration
What does gel electrophoresis do for the plasmid?
Measures the length of the protein
Why is supercoiled DNA the best for transient gene expression?
- Linear degrades
- Circular DNA can be damaged
- Supercoiled DNA is not vulnerable to degradation
What is the purpose of gold particle bombardment?
Using gold particles the DNA is directly delivered to the plant cells
What does CaCl2 do?
Neutralizes the charge associated with the DNA backbone and allows the coating of the DNA since both DNA and gold are negatively charged
What does spermidine do?
Precipitates DNA onto gold particles
What allows the gold particle bombardment to succeed?
When the gold particles travel at a fast velocity enter the nucleus to express the DNA
What happens after the transcription and translation of the gene?
The GFP is expressed and binds to the MAP4 encoding gene and eventually to the protein
