rate of enzyme controlled practical Flashcards
(12 cards)
which enzyme is used in this practical and why?
- trypsin, as it hydrolyses peptide bonds and produced in small intestine
what is the aim of the enzyme controlled reaction prac?
To investigate the effect of temperature on the rate of an enzyme-controlled reaction using trypsin and milk
what substrate does trypsin act upon?
casein, protein found in milk
whats the independent variable
temperature
dependant variables?
time taken for milke to become colourless, rate of reaction
control variables?
pH, enzyme conc, substrate conc, volume of reactants
main purpose of the water bath
maintain a constant temp for reaction
how to calculate rate of reaction?
1/time taken
why do we repeat?
to ensure reliability and account for anomalies
safety precautions?
gloves, goggles ti protect from trypsin, handle glassware accordingly
what is the expected trend in enzyme activity as temperature increases?
rate of reaction increases with temp, but beyond optimum enzyme may denature, so enzyme no longer works
how can you improve accuracy of timing
use colorimeter to measure change in absorbency as milk becomes colourless
