What is the preparation for the Microscopy practical
-Add a drop of water to a clean slide
-place the epidermal tissue into water on the slide
-Add a drop of iodine to stain
-place a cover slip on top of the specimen
What is the method for the microscopy practical
-clip the slide onto the stage
-ensure the lowest powered objective lens is over the slide
-use the coarse adjustment knob to bring the stage up just below the lens
-look down the eyepiece and gradually move the stage downwards using the coarse adjustment knob
-Adjust the fine adjustment knob to adjust the focus to make it clear
-change the objective lens to adjust magnification and readjust the stage using the coarse and fine adjustment knobs
What is the method for the growth practical
-wash hands and workspace
-use a permanent marker to divide the agar plate into sections
-soak a filter paper disk in your first antiseptic
-carefully place paper disk on one of the dots ensuring the lid is lifted away from your face
-soak paper disks in remaining antiseptics and add to the agar plate
-secure the lid of the plate with two small pieces of tape and incubate the plate
What is the method for the osmosis practical
-use a cork borer to cut 5 potato cylinders of the same diameter
-use a scalpel and ruler to trim each potato cylinder so they are all the same length
-measure the mass of each potato cylinder and record it in a table
-measure 10cm^3 of each sugar/salt solution and pour into each boiling tube
-Add one potato cylinder to each boiling tube and leave for a specified amount of time
-remove the potatoes
-blot dry and record the final mass and length of each
What is the method for the enzyme practical
-Place single drops of iodine solution in rows on the tile
-Use the syringe to place 2cm3 of amylase in the test tube
-Add 1cm3 of buffer solution to the test tube using a syringe
-Use another test tube to add 2cm3 of starch solution to the amylase and buffer solution
-start the stopwatch whilst mixing using a pipette
-After 10 seconds, use a pipette to place one drop of the mixture on the first drop of iodine, which should turn blue-black
-Wait another 10 seconds and place another drop of the mixture on the second drop of iodine
-Repeat every 10 seconds until iodine solution remains orange-brown
-Repeat experiment at different pH values

-Place a piece of pondweed into a beaker of water
-Use a light a set distance from the plant
-Record the number of bubbles observed in three minutes
-Repeat steps for different distances
