required pratical: effect of ph on amylase Flashcards
1
one drop of iodine solution into one well of the spotting tile
2
3 test tubes- 2cm cubed of starch, 2cm cubed of amylase solution, 2cm cubed ph 5 buffer solution
3
place all 3 test tubes in a water bath, leave for 10 minutes so they can reach right temp
4
combine all 3 solution into one test tube- mix with stirring rod, return to water bath and start a stopwatch
5
after 30 secs- use stirring rod to transfer one drop of solution in a well in the spotting tile that contains iodine
6
iodine should turn blue-black showing precence of starch
7
take sample every 30 secs - continue untill stays orange - tells us starch is no longer present
8
repeat experiment several times using different ph buffers
problem 1
taking sample every 30 secs - only have aprox time each reaction is complete- ever 10 secs
problem 2
looking for iodine not going blue-black- not obvious - gradual colour change - several people to look at when the reaction is complete
