Q

What did we expect for MIC for bleMBL and what happened?

A

We expected 16 in 0,4 and got the opposite

Q

What does the copy number have to say?

A

It has a lot to say for stability because of the division, if there is a high copy number the probability is high for every cell in the divison to get a copy.to ensure that every cell gets the plasmid has its systems

Q

what does the selection coefficient say?

A

represent the change in resistance over time
a positive selection coefficients means resistance will increase in prevalence over time. MSC itself is the concentration where the selection coefficient = 0. resistance will neither increase nor decrease over time.

Q

how do we calculate the selection coefficients?

A

first by calculating the ratios from the resistant/susceptible. the first graph was calculated by taking mean of the ratios from the replicates. then s(0-t48) was calculated from the generations. the coefficient was then calculated by taking the means of the s(0-t48)

Q

Why did the MIC strips for imipenem look so weird?

A

overexpression by both promoters and costly for them bith and thats why the growth is not confluent

Q

what is compensatory evolution

A

a hypothesis that even if a plasmid produce a cost when first introduced to a host the cost, could be alleviated over time through compensatory mutations in the plasmid or the host chromosome.
