What were your main two results?

A

1) AF senescent VSMCs in vessel wall LOWER in yes diagnosis of cSVD
- Mann Whitney p=0.0007
- But no significant difference in ratio

2) Higher ratio of senescent cells associated with LOWER cSVD score
- Spearman rank p=0.0480
- But no significant association fo AF

i.e. VSMC senescence is less frequently observed in cSVD arterioles than age matched healthy controls

How did your cSVD measures compare?

A

Yes diagnosis on PM of cSVD correlated with HIGHER cSVD score on histology

  • Mann Whitney p=0.0149
  • Increases confidence in both measures of cSVD

How did your senescence measures compare?

A

No significant association between ratio & AF on Pearson correlation

Ratio: assesses nuclear morphology & hence senescence

  • human interpretation, exclusions, assess tissue morphology & preservation
  • but vulnerable to bias and observer error

AF: computer generated quantification of abundance of heterochromatin

  • reproductible, quick, consistent, not subject to bias
  • but less specific as it didn’t exclude cells or lumen from calculations

How did other variables (than cSVD) compare to senescence?

A
  • Sex: a possible confounding factor - no significant difference on Mann Whitney
  • HTN: known risk factor - no signficant difference on Mann Whitney
  • Age: research says correlates with senescnece - no significant association found on Pearson correlation
