Q

steps of reticulin staining

A
  1. Oxidation (Potassium Permanganate)
  2. Bleaching (Oxalic Acid)
  3. Sensitization (Ferric Ammonium Sulphate)
  4. Silver Impregnation (ammoniacal silver)
  5. Reducing (Formalin)
  6. Toning (Gold Chloride)
  7. Rinsing (Sodium Thiosulphate)
  8. Counterstaining (Eosin)

Q

what are some precautions of reticulin staining

A
  • amoniacal silver is preapres fresh and very sensitive to pH
  • sensitization agen is tenuous and should not be subject to aggressive washing
  • unreduced silver left on slide will darken with time
  • high basic solutions may cause tissue to lift off slide
