Prevent normal physiologic release of norepinephrine from post ganglionic sympathetic neurons

Decrease of blood pressure due to inhibition of influence of sympathetic nervous system on heart and vessels

A

Arterial hypertension

Mechanism: blocks transport of norepinephrine to vesicles, causing its degradation by MAO and gradual depletion

Application and mechanism of guanethidine

A

Severe arterial hypertension resistant to other drugs
Mechanism : concentrated in vesicles, replacing norepinephrine. Hence, causes gradual depletion of norepinephrine and also inhibits release of norepinephrine to synaptic cleft

  • Orthostatic hypotension
