What is the function of the base of the microscope?
Contains the transformer (resp for voltage/energy output entering the lamp), the rheostat (controls light intensity), the illuminator (lamp).
What is the function of the mechanical stage?
- Support the specimen slides
- vernier scale = coordinates
What is the function of the slide holder?
- Hold the specimen slides on the stage and allows for smooth movements with the co-axial adjustment knobs.
What is the function of the stage adjustment knobs?
Move the slide in two direction to scan all parts of the slide.
What is the function of the field diaphragm?
Controls area of illumination
What is the function of the condenser?
Focus the light on the specimen
What is the function of the condenser adjustment knobs?
Moves the condenser up and down to focus the light directly on the specimen.
What is the function of the condenser centering screws?
Centers the condenser over the light.
What is the function of the iris diaphragm?
Adjusts the NA of the condenser to equal that of the objective in use. improves contrast.
What is the function of the objective lenses?
Responsible for primary magnification.
What is the function of the rotating nosepiece?
Holds and rotates objective lenses.
What is the function of the ocular lens?
- magnifies the image on the slide and must be adjusted to inter pupillary distance
What is the function of the diopter rings?
Used to focus the eyepiece for the left eye.
what is the function of the coarse and fine adjustment knobs?
Allows to move the stage up and down to focus the specimen under the objectives. Coarse = big adjustments fine= small fine adjustments.
What is resolution?
When two objects that are close together can be distinguished apart. More detail = better resolution.
What is magnification and what is total magnification?
ocular lens x Objective lens = TT magnification
What is the numerical aperture?
It’s the ability of the microscope to render the finest detail distinctly visible.
What is an empty magnification?
When the magnification has exceeded the resolving power and two distinct objects can no longer be distinguished. (Dumbbell)
What is definition?
The capacity of the objective lens to render the outline of an object as distinct.
What is spherical aberration?
is when many focal points produce a blurry image
What is the objective lens system?
-Responsible for primary magnification
-The ability to separate detail depends on the capacity of the lens for gathering light
What is coverslip glass?
Coverslips are little thin pieces of glass that adhere on top of a slide
purpose is to protect objective lenses and create even thickness.
What is slide glass?
It’s colourless, plain glass slides with a thickness between 0.9 to 1.1 mm thick
What is a plan-neofluar lens?
Contains fluorite which makes it idea for polarization microscopy due to its ability to transmit UV very well.
