A
  1. Binding to guanine bases of bacterial DNA –> block template function of DNA –> inhibit bacterial proliferation
  2. Alteration of membrane structure and its transport
  3. Disruption of mitochondrial electron transport chain

PK of Clofazimine

A

Absorption: 45-62% after oral administration (presence of food may increase rate and extent of clofazimine absorption)

Distribution: Primarily to fatty tissue and cells of reticuloendothelial system

Metabolism: Partially metabolized

Excretion: Partly found in feces (may represent excretion in bile)
