-using a special instrument to make sections (which are stained and mounted in a special chemical to preserve them)

how to find the actual size of the structures? eg a leaf in a transverse section

A

-measure the widest part of the leaf on the photomicrograph, in mm
-convert that measurement to µm, by multiplying by 1000x
-now divide this figure by the magnification (actual thickness of the leaf)
-use IAM formula to find magnification, if image is given
