How can wild strains of micro-organisms be improved?

What does recombinant DNA technology involve?
The use of recombinant plasmids and artificial chromosomes as vectors
What is a vector?
A DNA molecule used to carry foreign genetic information into another cell
What is the role of restriction endonuclease?
To cut open plasmids and specific genes out chromosomes, leaving sticky ends
What is the role of Ligase?
Seals a gene into a plasmid
What results in Mutations?
Exposure to UV light, Other forms of Radiation OR Mutagenic chemicals
Why are Artificial chromosomes preferred over plasmids as vectors in some circumstances?
When are complementary sticky ends produced?
when the same restriction endonuclease is used to cut open the plasmid and the gene from the chromosome
What do restriction sites contain?
they contain target sequences of DNA where specific restriction endonuclease cuts
What does a regulatory sequence do?
it controls gene expression
What does the origin of replication allow?
It allows self-replication of the plasmid/artificial chromosome
What do selectable markers do?
They protect the micro-organisms from a selective agent (antibiotic) that would normally kill it or prevent it growing
