18
Q

1st step in Smear and Staining?

A

Smear Preperation

19
Q

Describe Smear Prepration

A

Drop a droplet of water onto a slide. Add specimen and mix together.

20
Q

2ns step in Smear and Staining?

A

Fixing.

21
Q

Describe Fixing

A

Fixing is heating the specimen to “fix” it to the slide. Can be done with heat or alcohol or ethanol or acetone.

22
Q

Simple Stain tells you what?

A

Only tells shape. Not very useful

23
Q

Differential stains are either?

A

Acid Fast or Gram Stain

24
Q

Basic Dyes (+ ions)

A

crystal violet, methylene blue, **safranin (pink), carbol fuchsin (red)

25
Q

Acidic Dyes (- ions)

A

India Ink, Negrosin, Eosin

26
Q

What type of stains distinguish bacteria?

A

Gram or Acid Fast

27
Q

Gram stains tell you what about cell wall?

A

thickness

28
Q

Gram + has _______ cell wall

A

thick

29
Q

Gram - has _______ cell wall

A

thin

30
Q

Gram Stain Procedure (9 steps)

A
  1. Smear Prep
  2. Heat fix or alcohol fix (5min)
  3. Add Crystal Violet 1-3min
  4. Rinse in water
  5. Add Gram’s Iodine 1-3min
  6. Rinse in water
  7. Decolorize w/ 95% alcohol
  8. Add Saffranin (counter stain) 2min
  9. Rinse in water.
31
Q

Gram stain Procedure dyes, in order

A

Crystal Violet, Gram’s Iodine, Decolorize with 95% alcohol, saffranin

32
Q

Antibiotic selection for Gram + (2 of them)

A

Penicillin and sulfonamides

33
Q

Antibiotic selection for Gram - (3 of them)

A

Streptomycin, tetracycline, chloramphenicol

34
Q

Acid Fast Stain to view ______ and ________

A

Mycobacteria and Nocardia

35
Q

Micro bacteria and Norcardia have _______ content of lipids.

A

High. 20-40% lipids in cell wall.

36
Q

Tuberculosis should be viewed using what stain?

A

Acid Fast Stain

37
Q

Acid Fast Stain Procedure (8 steps)

A
  1. Smear Prep
  2. Heat fix. 80degrees for 20min
  3. Add Carbol fuchsion (red) and heat* (slide over heat gently 1-3 min. Heat melts lipids.
  4. Cool and rinse with water.
  5. Decolorize with acid-alcohol.
  6. Rinse in water
  7. Counterstain with methylene blue
  8. Blot dry - examine
38
Q

Acid fast stain bacteria what color?

A

Red

39
Q

Non-acid fast stain what color?

A

Blue
