cryobiology Flashcards

1
Q

What is vitrification?

A

defined as glass like solidification of supporting solutions and living cells that completely avoids ice crystals formation during cooling and warming

Q

Why are cryopreservation procedures done?

A

this is done to…

  1. reduce risk of microbial contamination
  2. reduce risk of cross contamination with other cell lines

Q

What is the difference between controlled cooling and uncontrolled cooling

A

controlled cooling=(1C/min), requires controlled rate freezer, which is expensive

uncontrolled cooling-freezer, followed by -80C overnight before transferring to liquid nitrogen for storage

Q

What is the purpose of cryoprotectants

A

to minimize/ prevent cells/tissue or organ from being damage during exposure to low temperature.

forms hydrogen bond to prevent water-water hydrogen bonding that results in ice formation

Q

State four ways that a cell culture system can get contaminated

A
  • poor handling
  • from contaminated media, reagents, and equipment
  • from microorganisms present in incubators, refrigerators, and laminar flow hoods
  • from skin of the worker and in cultures from other laboratories
