Antiepileptic Drugs Flashcards
Two or more recurrent unprovoked seizures.
Epilepsy
What are the types of seizures?
1-Partial seizures.
Simple/complex
2- Generalized seizures.
Inhibits GABA A receptors activity.
Bicuculline/ Picrotoxin
Agonists at glutamate receptors.
Kainic acid ( result in excitotoxic damage)
What is the first line drug used for partial seizures?
Carbamazepine
Used as second line treatment of partial seizures?
Tiagabine, valproate, gabapentin
Used as first line treatment of generalized tonic-clonic seizures.
Carbamazepine, valproate
Second line treatment for generalized tonic-clonic seizures.
Phenytoin, phenobarbitone
First line treatment of absence (petit mal) seizures.
Valproate, ethosuximide
Second line treatment of absence seizures.
Clobazam, clonazepam
How does anti-epileptic drugs act on GABA receptors?
Either to
1- Inhibit GABA transaminase (responsible for metabolism of GABA).
2- or block the re-uptake by blocking the transporters.
What are the drugs used for inhibiting GABA transaminase activity?
V.V
Valproate, Vigabatrin
What does tiagabine do to increase GABA levels?
Blocking the re-uptake of GABA
GABA A agonists.
Barbiturates, benzodiazepines
Modulate the activity of GABA by increasing the frequency of Cl channel opening.
Benzodiazepines
