What are bisphosphonates?
Bisphosphonates are analogs of pyrophosphates that specifically inhibit bone resorption.
What condition are bisphosphonates effective in treating?
They are very effective for the treatment of postmenopausal osteoporosis.
Where do bisphosphonates concentrate in the body?
Bisphosphonates concentrate at sites of active remodeling.
What happens to bisphosphonates during bone remodeling?
They remain in the matrix until the bone is remodeled, then they are released in the acid environment of the resorption lacunae.
What effect do bisphosphonates have on osteoclasts?
They induce apoptosis in osteoclasts.
How are bisphosphonates classified?
They are classified mainly based on potency.
What are the first-generation bisphosphonates?
First-generation bisphosphonates include Etidronate (least potent), Clodronate, and Tiludronate.
What are the second-generation bisphosphonates?
Second-generation bisphosphonates include Alendronate, Pamidronate, and Ibadronate, which are 10-100 times more potent than the first generation.
