Propylthiouracil (PTU)
- Inhibits thyroid peroxidase → ↓ T3/T4 synthesis
- Also blocks peripheral T4 → T3 conversion
Methimazole
Inhibits thyroid peroxidase → ↓ T3/T4 synthesis
Amiodarone
- Can inhibit peripheral conversion of T4 → T3
- Contains iodine → may cause hypo/hyperthyroidism
Beta-blockers
- Inhibit T4 → T3 conversion (some types)
- Used in thyrotoxicosis
Lithium
Inhibits thyroid hormone release
Thiocyanates / Perchlorate / Nitrate / Goitrin
Compete with iodide uptake → inhibit hormone synthesis
Sulfonylureas, Glybenclamide
Stimulate insulin release from β-cells (block K⁺ channels)
