Practical on investigation of how ph affects enzyme activity
- Put a drop of iodine solutions into every well of spotting tile
- Set up water bath at 35°C
- Add amylase solution and buffer solution in pH 5 into boiling tube
- Put boiling tub in the water bath and wait 5 mins
- add starch solution to boiling tube, mix and start stop clock
- Every 10 seconds take a sample from boiling tube using a dropping pipette
- Put a drop of sample into well on spotting tile
- When iodine solution stays browny-orange all starch in sample has been broken down
- Repeat experiment with buffer solution of different pHs
As the pH changes what else will change?
The time it takes for starch to be broken down
What should stay the same to make it a fair test?
