Captopril, rampiril in high BP, HF and chronic renal insufficiency
What is Km?
Substrate conc where reaction rate reaches half of Vmax
Competitive inhibitor?
Competes with substrate for active site
Reversible inhibitor
Overcome by increase in substrate, Km up but Vmax same
Irreversible inhibitor
Covalent binding, renders active site unusable, decreases Vmax
Example of irreversible inhibitor
Selegiline, irreversible inhibitor of MAO B

Produces false transmitter, alpha methyl dopa

Inducible by injury or inflammation

Cox 1 and 2, lasts until new enzyme synthesised
How do sulphonamides work?
Compete with PABA for uptake in bacteria, prevent DNA synthesis by inhibiting dihydrofolate synthase
