What is the practical purpose of the aseptic technique described?
To grow bacteria from the room we are working in.
What apparatus is needed for the experiment?
1 petri dish, 1 bottle of warm, fluid agar, Sellotape, cotton buds, 1 Bunsen burner, heatproof mat, safety glasses.
What is the first step in the method?
Set up the Bunsen burner on the heatproof mat.
How should the petri dish be prepared?
Place the petri dish on the mat close to the Bunsen and draw a cross on the outside of the base to divide it into 2 equal areas.
What should be done with the agar bottle before pouring?
Carefully unscrew the lid and flame the mouth of the bottle in a blue Bunsen flame.
What is the procedure for pouring the agar into the petri dish?
Lift the lid of the petri dish just enough to pour the agar in, then close the lid as quickly as possible.
What should be done after pouring the agar?
Gently move the dish to ensure the bottom is completely covered in agar and allow 5 - 10 minutes for it to set.
What surfaces can be chosen to obtain microbes?
Suggestions include the surface of your desk, door handle, sole of your shoe, iPad screen, keyboard, rim of bin, tap, etc.
How should microbes be collected from the chosen surface?
Use a cotton bud to obtain microbes from the chosen surface.
What is the final step in transferring microbes to the agar?
Return to the work area, lift the lid of the petri dish, and stroke the cotton bud over the surface of the agar without damaging it.
