LAB Flashcards
Toluidine Blue
Stains cellulosic Walls purple and lignified walls light blue. Is a good general purpose stain.
Aniline Blue
Stains nuclei, cytoplasm and callose blue
phlorogucinol/HCL
PREPARE A 1:1 SOLUTION IN A SMALL PETRI DISH AND FLOAT SECTIONS FOR 2-5 MINUTES FOR STAINING. stains lignified walls red.
IKI
Stains starch grains dark blue; cellulose walls brown; cork, cutin and callose yellow.
Ruthenium Red
Stains pectins red.
Clearings
NaOH is used to remove the cellular contents to make clearings. After treatment only the cell walls remain. Clearings are commonly used to examine patterns of cell division, epidermal structures and patterns of vascular tissue.
Macerations
Tissue treated with acid or base which dissolves the middle lamella (glue between cell walls) and causes the individual cells to come apart allowing them to be viewed individually.
Root System
Tap root, lateral roots, root hairs
Shoot system
cotyledons, hypocotyl, stem (nodes and internodes); foliage leaves (base, petiole and lamina); Leaf axil; axillary buds; shoot apex; leaf primordia (youngest leaves)
Helianthus
Sunflower
