lab 8 Flashcards
(35 cards)
What is a gelatin test?
it tests for PATHOGENICITY of bacteria and if bacteria have enzyme gelatinase present.
What does positive gelatin test indicate?
liquified
what does negative gelatin test mean?
solid
What does catalase test do?
-checks for catase.
If oxygen is reduced, the production of superoxide radical can occur.
CATALASE destroys radicals.
Positive catalase =
bubbles
negative catalase =
no bubbles
What does coagulase test do?
coagulase is present if bacteria can form a clot around themselves for protection. Positive test is CLOTTING.
What does DNase test do?
DNase can degrade a host’s DNA and show PATHOGENICITY of bacteria.
what does positive test for DNase indicate ?
clear zone around colony
What is oxidase test for?
-If oxidase enzyme is present as significant for energy production. used in ETC
what is positive test for oxidase ?
purple color
negative test for oxidase ?
light pink
What is urease test for?
to break down urea and in forming ammonia. Ammonia causes rise in pH.
positive test for urease is
pink/purple color
negative test for urease is
yellow/green color
what is cysteine test for ?
to identify is cysteine desulfurase is present. sulfur will be removed and H2S gas present
what is positive result of cysteine test ?
black precipitate
what is negative result of cysteine test ?
no precipitate
Phenylalanine Deamination Test
Checks for phenylalanine deaminase and presence of pyruvic acid.
-adding ferric chloride to test if present
Positive Phenylalanine Deamination Test indicates ..
green color
negative Phenylalanine Deamination Test indicates
yellow
What is nitrate reduction test?
to check for nitrate reductase
PRODUCT NEEDS TO BE NITRITE
What indicates if nitrite is present ?
IF REDUCES
with addition of sulfanilic acid and dimethyl naphthylamine- the ions produce a red color change
What if no color change?
No reaction occurred or may been reduced by another ENZYME
