What is a pathogenic organism?
disease causing
what is an opportunistic pathogen?
not normally disease causing but when immune is low will cause disease.
what is free living organism?
lives freely in the environment and not disease causing.
why are environmental streaks different from quadrant streaks on plate? (3)
- less bacteria then a clinical sample
- with less bacteria quadrant streaking is ineffective.
- quadrant streaking is difficult in the field with out access to a flame.

what is agar?

What is agar used for in TSA plates?
used as a solidifier/ gelling agent for TSA and other media
why is agar better to use then gelatin?
agar is firmer and is not degraded by bacteria.
what are the 3 parameters to sterility? (autoclave)
- temperature
- time
- pressure
Why are the 3 sterility parameters so important?
only way to kill fungus spores.
what are the 6 cultural characteristics used to identify colonies on a plate?
size, color, texture, elevation, form, margin
what 4 characteristics are used to describe broths?
surface, amount of growth, color, subsurface,
why is staining bacteria important?
bacteria are nearly invisible without staining.
what are 3 common positively charged stains?
Methylene blue, safranin, crystal violet
what are 2 common negative stains?
Eosin, acid fuschin
how does a stain work? (2 parts)
- Chromophore that colors the bacteria and binds to the Auxochrome
- Auxochrome that binds to the bacteria.
