7
Q

what are the three tenets of cell theory?

A

1) all living organisms are composed of one or more cells
2) the cell is the basic unit of structure and organization in all organisms
3) cells arise from preexisting cells

16
Q

what did Golgi get the nobel prize for?

A

for the dark reaction

17
Q

how is the dark reaction staining done?

A
  1. fix the sample with formaldehyde
  2. put the sample in silver nitrate for 2 days
  3. cut tissue sections
  4. dehydrate the sample using ethanol
  5. mount coverslip
  6. look on microscope
  7. microcrystallization of silver chromate in the cell
18
Q

what lead to the creation of the neuron theory?

A

fine tuning the dark reaction with gold instead to get finer details

19
Q

who coined the term neuron and who came up with the neuron theory?

A

-term coined by Heinrich Wilhelm
-theory made by Santiago Cajal (godfather of neural science)

20
Q

which company patented the electron microscope and which two people made the electron microscope?

A

-Siemens company patented it because Rudenberg (a member of the company) wanted to see the polio virus that infected his son
-Ruska and Knoll made the first electron microscope

21
Q

how much better was the electron microscope compared to the light microscope?

A

-light microscope had a magnification of 100x and a resolution of 200nm
-electron microscope have a 12,000 mag and 50nm

22
Q

who made the first cell line in 1952? what cells did he use?

A

George Otto Gey, used Henrietta Lacks cervical cancer cells (HeLa, no consent)

23
Q

what is the backstory behind the first cell line?

A

-Henrietta Lacks was in the hospital for cervical cancer
-a tissue sample was removed from her and sent to Dr. Gey
-he tried to make the cell line but failed after a few tries
-Mary Kibicek put the tissue sample in a roller tube and they grew well

24
Q

how did we get the first cell line?

A

because the tissue was cancerous, grew fast, and grew generation after generation

25
who created the first transgenic mice in 1974?
-Rudolph Jaenisch, used DNA virus to infect an early mice embryo, the cells contained the viral DNA making it transgenic but it couldn't be passed on through the germ line -this made it not so useful
26
which people mutated the germline in mice to allow the viral DNA to be passed on in 1981?
-Frank Ruddle -Elizabeth Lacy -Ralph Brinster -Richard D Palmiter
27
who discovered Green fluorescent protein (GFP) in 1960-1970?
Osamu shimomura
28
what did Douglas Prasher propose? why did he give his work out?
that GFP can be used as a molecular tracer, because he couldn't get a grant for his work
29
why did the cDNA (copy DNA) of GFP gene not work when Prasher gave it out?
it was still associated with jellyfish DNA
30
what did Martin Chalfie do?
expressed GFP without jellyfish amino acids in E. coli and C. elegans
31
who also expressed GFP in E. Coli but was a month late?
Frederick Tsuji
32
who discovered the mutated GFP with different properties and who made the different colors for it?
-Roger Tsien
33
what is super resolution microscopy?
techniques to go beyond the resolution of a microscope of 0.2 nm
34
who came up with STORM?
Xiawei Zhuang
35
who came up with PALM?
Eric Betzig
